Malaria vaccines in the future--what strategies now?

چکیده

In addition to the inherent challenge, driven by curiosity and the excitement of the chase, the quest for malaria vaccines has been made more imperative over the past two decades or so by pragmatic assessment of the odds in a disease which has been with mankind for millenia. Accel erating spread of parasite resistance to more and more drugs, with too few new molecular structures in the pipeline casts a dark shadow over the future the immediate future. Coupled with insecticide resistance of vectors and the momentous cost in money and human effort absorbed by classical malaria control programs, the pressure of decreasmg drug efficacy became critical in making available a relative profusion of funds over a short time period for the vaccine quest on a global scale.

Need for Optimisation of Immunisation Strategies Targeting Invasive Meningococcal Disease in the Netherlands

Invasive meningococcal disease (IMD) is a severe bacterial infectious disease with high mortality and morbidity rates worldwide. In recent years, industrialised countries have implemented vaccines targeting IMD in their National Immunisation Programmes (NIPs). In 2002, the Netherlands successfully implemented a single dose of meningococcal serogroup C conjugate vaccine at the age of 14 months a...

Malaria genome project and its impact on the disease.

Malaria remains uncontrolled to-date due to lack of effective parasite and vector control strategies. With the completion of the host, parasite and vector genome projects more suitable and effective disease control measures can be achieved. Here we have reviewed the Plasmodium falciparum genome project and its impact on malaria research in future. The parasite genome project has revealed certai...

Strategies for Designing and Monitoring Malaria Vaccines Targeting Diverse Antigens

After more than 50 years of intensive research and development, only one malaria vaccine candidate, "RTS,S," has progressed to Phase 3 clinical trials. Despite only partial efficacy, this candidate is now forecast to become the first licensed malaria vaccine. Hence, more efficacious second-generation malaria vaccines that can significantly reduce transmission are urgently needed. This review wi...

Pregnancy-associated Malaria and the Prospects for Syndrome-specific Antimalaria Vaccines

Aided by the Plasmodium falciparum genome project, recent discoveries regarding the molecular basis of malaria pathogenesis have led to a better understanding of the interactions between host and parasite. Although vaccines that prevent infection by malaria parasites remain only hopes for the future, there are now more immediate prospects for vaccines that protect against specific disease syndr...
